LIPASES. Expert has spent most of his scientific career studying the regulation of lipases and other enzymes which function at oil-water interfaces. The emphasis of his work has been placed upon studying the role of lipid-lipid and lipid-protein interactions in the control of lipolysis at the molecular level. Lipids with which he has been particularly interested are lecithin, diacylglycerol, fatty acid, and cholesterol.

CHOLESTEROL ESTERASES. As a part of his study of lipases in general, Expert has devoted considerable effort to the study of cholesterol esterases. He notes that these are important enzymes in mammalian cholesterol homeostasis. The mammalian form most studied by Expert is that which belongs to the cholinesterase gene family. This is also known as bile-salt-stimulated lipase, lysophospholipase, carboxylester lipase, sterol ester hydrolase, and non-specific lipase.

ADSORPTION. Expert has extensive expertise in studying enzyme, peptide and drug adsorption to lipid-water interfaces. He notes that this has been necessitated by his lipase interest because lipases must adsorb to the lipid-water interface before they can act on substrates in it. The emphasis of his work on adsorption has been the retention of catalytic activity. Examples of this include the lack of protein denaturation and the role of the 2-dimensional organization of lipids in regulating protein adsorption. His recent focus has been on the application of percolation theory to the regulation of lipase adsorption and catalysis.

MONOMOLECULAR FILMS. Expert has made extensive use of monolayer techniques as a model lipid-water interface for studying lipase adsorption and catalysis. For analyzing such data, he has developed mathematical algorithms to characterize the surface potential-surface pressure-area characteristics of mixtures of lipids in monomolecular films. Lipids of interest to him have been phospholipids, acylglycerols, fatty acids, and cholesterol esters. Expert has developed automated instrumentation to facilitate the study of lipid-lipid and lipid-protein interactions in monomolecular films. He has emphasized the detection and elimination of surface-active impurities in this work. His goal in doing so has been to insure the accuracy and precision of his data.
