Chloroperoxidase, an enzyme which uses H2O2 and catalyzes halogenation and epoxidation reactions, was discovered in Expert's laboratory many years ago. Subsequent research has involved studies on the mechanisms of the reactions and the utility of chloroperoxidase in the synthesis of chiral epoxides. The gene for chloroperoxidase has been cloned. Current efforts are devoted to site specific mutagenesis in order to extend stereoselectivity to halogenation reactions and to expand the number of organic acceptor molecules which function in halogenation and epoxidation reactions. Using "directed evolution" techniques the Expert lab has produced chloroperoxidase mutants that can function in the presence of high concentrations of organic solvents. A second directed evolution mutant is resistant to mechanism based inactivation during the epoxidation of primary olefins.

Halophytic plants synthesize and distill methyl chloride into the atmosphere. Expert's interest has been "how and why" plants carry out this reaction. He has isolated and cloned an enzyme which catalyzes the transfer of a methyl group from S-adenasye methionine to chloride ion. This enzyme answers the question "how." He is attempting to answer the question "why." His basic hypothesis is that halophytic plants control their internal chloride ion concentrations by making a volatile methyl chloride. He is attempting to clone the transferase gene and transform soybean plants to enable them to make methyl chloride. He the plans to compare wild-type and transformed soybeans in their ability to grow under high salt environments.
