MEMBRANE TRAFFIC. All of Expert's work in the last ten years has involved membrane traffic, ranging from genetic analysis of endocytosis and secretion in mammalian cells to biochemical analysis of the entry of protein toxins, including immunotoxins, into cells. HYBRID TOXINS; IMMUNOTOXINS. The goal of Expert's work with protein toxins is the construction of more potent and specific hybrid immunotoxins that may have chemotherapeutic value. He and his colleagues have applied to patent an approach in this area using toxins based on Pseudomonas aeruginosa exotoxin A. ENDOCYTOSIS. Expert has done extensive work on the endocytosis of protein toxins and on the endocytosis of transferrin and the transferrin receptor. He has also worked a great deal on mammalian cells expressing genetic defects in the endocytic pathway. SECRETION. Over the last four years, Expert has studied secretion as it relates to mutants he has isolated that are defective in the secretory pathway. PSEUDOMONAS AERUGINOSA EXOTOXIN A. For the last five years, Expert has been studying the molecular and cellular biology of the Pseudomonas aeruginosa extoxin A, especially ways to improve immunotoxins made with exotoxin A. He and his colleagues submitted a patent application covering some of this work. DIPHTHERIA TOXIN. Although the diphtheria toxin is not currently his focus, Expert has had a lot of experience with this toxin in the past and continues to follow the literature in the field.
